目的 探讨高原低氧对大鼠大脑皮质生长休止蛋白7(Gas7)表达的影响.方法 36只大鼠随机分为正常对照组和模拟高原低氧组,模拟高原低氧组大鼠进行6周缺氧,复制慢性高原低氧动物模型.实验结束后,所有动物采用免疫组织化学和免疫印迹技术检测大鼠大脑皮质中Gas7的表达.结果 与对照组相比,Gas7在模拟高原低氧组大鼠大脑皮质的表达明显增强.结论 Gas7可能参与了高原低氧对大鼠大脑皮质神经元结构和功能的影响.  相似文献

南海北部陆坡神狐海域HS-PC500 岩心微生物多样性   总被引:3,自引:0,他引:3  
[目的]本文研究南海北部陆坡神狐HS-PC500重力活塞岩心沉积物中微生物多样性.[方法]使用吖啶橙染色法计数沉积物中微生物丰度;提取沉积物微生物总DNA,使用特异性引物扩增古菌及细菌16SrRNA基因序列;对克隆文库进行系统发育分析.[结果]系统发育分析显示表层PC500-l(0-5 cm below sea floor,bsf)古菌以C3为主要类群,占该层总序列的25.6%;中层PC500-6(350-355 cm bsf)和底层PC500-11(790-795 cm bsf)古菌以Marine Benthic Group(MBG)-B为主要类群,分别占该层总序列的48.1%和38.9%.另有部分克隆序列属于MBG-A、Miscellaneous Crenarchaeotic Group(MCG)、Thermoprotei、NGC、Halobacteriales、MBG-E、South African Gold Mine Euryarchaeotic Group(SAGMEG).表层细菌以变形菌(Proteobacteria)为主要类群,占该层文库的38.3%.中层和底层细菌以绿弯菌(Choloflexi)和JS1为主要类群,分别占该层文库的28.1%、29.2%和39%、24.7%.另有部分克隆序列属于硝化螺旋菌(Nitrospirae)、放线菌(Actinobacteria)、酸杆菌(Acidobacteria)、OP8、螺旋体菌(Spirochaetes)、TM6、脱铁杆菌(Deferribacteres)、浮霉菌(Plantomycete).[结论]结果显示,HS-PC500岩心微生物丰度与甲烷浓度变化相吻合;微生物丰度较低可能与较低的总有机碳量有关;微生物多样性较高,并且随深度的增加群落结构变化明显;岩心中有关硫酸盐还原的微生物类群占优势,说明微生物的硫代谢在该海区沉积物的物质循环过程中占有重要地位.  相似文献

目的探讨生长休止蛋白7(Gas7)在成年大鼠关节软骨中的表达及意义。方法雄性SD大鼠(3个月、12个月)各12只。取膝关节和股骨上端关节部分,经脱钙石蜡包埋并切片,采用HE染色和免疫组化SABC法检测。检测各关节软骨组织中Gas7的表达及其定位。结果关节软骨切线层与移行层Gas7的表达呈现强阳性,而在辐射层及软骨钙化层则弱阳性或阴性。结论Gas7在成年大鼠关节软骨各层细胞的表达不同,可能与各层软骨细胞的生长发育及代谢状况有关  相似文献

In industry, fosfomycin is mainly prepared via chemical epoxidation of cis-propenylphosphonic acid (cPPA). The conversion yield of fosfomycin is less than 50% in the whole process and a large quantity of waste is produced. Biotransformation by microorganisms is an alternative method of preparation. This kind of conversion is more delicate, environmentally friendly, and the conversion yield of fosfomycin would be higher. In this work, an aerobic bacterium capable of transforming cPPA to fosfomycin was isolated. The organism, designated as strain S101, was identified as Bacillus simplex by morphological and physiological characteristics as well as by analysis of the gene encoding the 16S rRNA. Fosfomycin was assayed by two means, bioassay and gas chromatography (GC). Glycerol was a good carbon source for growth and cPPA conversion of strain S101. When cPPA was used as the sole carbon source, neither growth nor conversion to fosfomycin occurred. The optimum cPPA concentration in the conversion medium was 2,000 μg ml−1. After 6 days of incubation, the concentration of fosfomycin reached its maximum level (1,838.2 μg ml−1), with a conversion ratio of 81.3%. Air was indispensable for the growth but not for the conversion to fosfomycin. Furthermore, vanadium ions were found to be essential for the conversion. High concentrations of cPPA had fewer inhibitory effects on the growth of strain S101.  相似文献

Moderate heat stress has been reported to increase PSI cyclic electron flow (CEF). We subjected leaves of Arabidopsis (Arabidopsis thaliana) mutants disrupted in the regulation of one or the other pathway of CEF flow—crr2 (chlororespiratory reduction, deficient in regulation of chloroplast NAD(P)H dehydrogenase-dependent CEF) and pgr5 (proton gradient regulation, proposed to have reduced efficiency of antimycin-A-sensitive-CEF regulation) to moderate heat stress. Light-adapted leaves were switched from 23 to 40°C in 2 min. Gas exchange, chlorophyll fluorescence, the electrochromic shift (ECS), and P700 were measured. Photosynthesis of crr2 and pgr5 was more sensitive to heat and had less ability to recover than the genetic background gl. The proton conductance in light was increased by heat and it was twice as much in pgr5, which had much smaller light-induced proton motive force. We confirmed that P700 becomes more reduced at high temperature and show that, in contrast, the proportion of PSII open centers (with Q A oxidized) increases. The two mutants had much slower P700+ reduction rate during and after heat than gl. The proportion of light absorbed by PSI versus PSII was increased in gl and crr2 during and after heat treatment, but not in pgr5. We propose that heat alters the redox balance away from PSII and toward PSI and that the regulation of CEF helps photosynthesis tolerate heat stress.  相似文献

In this study, a modified version of the gas production technique was used to determine protein fermentation characteristics in rumen fluid of 19 feedstuffs. Performing the incubations in a N-free environment, and with an excess of rapidly fermentable carbohydrates, made N the limiting factor to microbial growth, and so gas production profiles reflected the availability of N from the feed samples. Results showed that fermentation of protein in rumen fluid can be determined with this modified gas production technique, and that there were distinct differences in protein fermentation between the feed samples. Availability of protein for fermentation was highest in wheat, potato pieces and lupin, and lowest in Rumiraap, a formaldehyde treated rapeseed meal, palm kernel expeller and brewery grains. The protein degradation characteristics of the 19 feed ingredients were also determined with the in situ nylon bag technique. With the obtained results, the amount of rumen escape protein (REP) was calculated for each feedstuff. The results showed that the rate of degradation ranged from 0.010/h for Rumiraap to 0.151/h for wheat. The amount of REP ranged from 197 g/kg CP for lupin to 840 g/kg CP for Rumiraap. Comparing the gas production results with the results obtained with the nylon bag technique showed that there was a good relationship between the gas production after 12–25 h of incubation and the calculated amount of REP (r2 = 0.83–0.85). The results show that the adapted gas production technique, being depleted of N and using an excess of rapidly fermentable carbohydrates, is suitable to recognize differences in N availability between feed samples and can be used as an alternative to the nylon bag technique and other in vitro techniques.  相似文献

Introduction – Ganoderma, one of the best‐known traditional Chinese medicines, has attracted considerable attention owing to the fact that dozens of polysaccharides isolated from it have shown diverse and potentially significant pharmacological activities. However, no work has been reported on the analysis of monosaccharide composition of polysaccharide isolated from the aqueous extract of Ganoderma atrum yet. Objective – To develop a simple and sensitive GC‐based method for the analysis of monosaccharide composition of purified polysaccharides in Ganoderma atrum. Methodology – The polysaccharide was first hydrolysed to give the constituent monosaccharides, which were subsequently derived into acetylated aldononitriles and analysed by gas chromatography using a capillary column packed with a (5%phenyl) methylpolysiloxane stationary phase with the addition of acetyl inositol as the inner standard. High‐performance liquid chromatography was also used for comparison. Results – The stable derivatives of the most common monosaccharides could be separated and reproducibly determined with high sensitivity. The limits of detection and quantification were 0.013 and 0.043 mg/mL, respectively. The intermediary precision values (expressed as the RSD) were less than 10%. The mean recovery of the method was 100 ± 3%, with RSD values of less than 5%. The results obtained from GC and HPLC methods were found to be close to each other within acceptable error ranges. Conclusion – This study demonstrated that the developed method could be applied as an accurate method for the compositional analysis of monosaccharides in the field of biological and biochemical study. Errors that are commonly made in the quantification of biogas from anaerobic digestion experiments were investigated.For liquid displacement gasometers where a barrier solution separates the biogas and the atmosphere, inaccuracy due to gas diffusion was examined experimentally. Acidified saturated saline solution was the most suitable barrier solution, as biogas characteristics changed least with time. Using acidified or tap water caused considerable biogas losses and should therefore be avoided where biogas is stored before measurement.Errors associated with volume calculation from three common liquid displacement gasometer types were investigated theoretically. Corrections that must be made to obtain gas volumes at standard temperature and pressure when using this equipment are discussed. Regarding experimental errors, gasometer designs where displaced liquid is weighed to determine the volume are the most versatile since errors depend mainly upon balance sensitivity. Using liquid heights to calculate volume requires appropriate sizing of the gasometer relative to the volume of gas measured.The calibration of a low flow gas meter was investigated and an approximately linear variation with flow rate was found; hence in situ calibration is advised for this type of instrument. Correction for atmospheric conditions should be performed in real time to reduce errors.  相似文献

如何提高产气量一直以来都是沼气发酵过程中需要解决的主要问题.本研究是在实验室条件下组建模拟沼气发酵罐,并用于研究发酵罐中添加尿素对沼气发酵的影响.结果表明:与不添加尿素的对照组相比,处理组中添加浓度为1 000 mg/L的尿素可以有效提高沼气发酵基质的利用率,并提高沼气中甲烷含量,该系统中发酵原料总固体(total solid,TS)消耗率提高1.79%,挥发性固体(volatile solid,VS)消耗率提高4.36%,甲烷产量提高8.4%;添加3 000 mg/L尿素使发酵原料TS消耗率降低1.59%,但是VS消耗率提高2.28%,产甲烷量提高5.4%;添加5 000 mg/L尿素则会对发酵原料的利用产生抑制作用,TS、VS消耗率分别降低3.04%、3.72%,甲烷产量降低4.2%.本研究结果将为提高沼气发酵效率提供初步的理论基础.  相似文献
